Takara The NucleoSpin 96 DNA FFPE kit is designed for high-throughput DNA purification from formalin-fixed, paraffin-embedded samples, employing an odorless, xylene-free paraffin removal technology. After the paraffin is dissolved, Proteinase K digestion is used to solubilize the fixed tissue and release DNA into solution, and the samples are heated in an optimized buffer to remove crosslinks. DNA is then bound to the NucleoSpin silica membrane in the presence of ethanol and chaotropic salts, followed by two washing steps and elution of purified DNA under low ionic strength conditions in a slightly alkaline buffer.
