Background Information | Endostatin is a proteolytic cleavage product from the C-terminus of Collagen XVIII, which is itself a proteoglycan/collagen found in blood vessel walls and in basement membranes. Endostatin is a compact globular fold-containing protein with a Zn2+ binding site near its amino terminus. Originally identified in conditioned media of murine hemangioendothelioma (EOMA) cells, endostatin is generated from the parent collagen XVIII molecule involving intermediate glycosylated forms and the proteolytic activity of proteases such as cathepsin L, metalloproteases, and elastase. Endostatin has a wide-spread tissue expression and has been shown to co-localize with fibulins, nidogen-2 in basement membranes, and elastic fibers of veins. Endostatin has also been shown to cause a G1 arrest in endothelial cells and to induce apoptosis. |

UniProt Summary | FUNCTION: Endostatin potently inhibits endothelial cell proliferation and angiogenesis. May inhibit angiogenesis by binding to the heparan sulfate proteoglycans involved in growth factor signaling. SUBCELLULAR LOCATION: Secreted › extracellular space › extracellular matrix (By similarity). TISSUE SPECIFICITY: Expressed in liver, kidney, lung, skeletal muscle and testis. PTM: Prolines at the third position of the tripeptide repeating unit (G-X-Y) are hydroxylated in some or all of the chains. SEQUENCE SIMILARITIES: Belongs to the multiplexin collagen family. Contains 1 FZ (frizzled) domain. Contains 1 TSP N-terminal (TSPN) domain. |
Molecular Weight | ~28 kDa observed. A ~28 kDa endostatin fragment has been observed in western blot as a result of MMP-7 cleavage of collagen XVIII (Chang, J., et al. (2003). Invest Ophthalmol Vis Sci. 44:832). |
