Background Information | Cytochrome P450 26B1 (UniProt Q9NR63; also known as Cytochrome P450 26A2, Cytochrome P450 retinoic acid-inactivating 2, Cytochrome P450, subfamily XXVIB, polypeptide 1, Cytochrome P450RAI-2, Retinoic acid-metabolizing cytochrome) is encoded by the CYP26B1 (also known as CYP26A2, P450RAI2, RHFCA) gene (Gene ID 56603) in human. Cytochromes P450 (CYP) proteins are primarily membrane-associated oxidases located either in the inner membrane of mitochondria or in the endoplasmic reticulum. In addition to processing endogenous substrates, CYPs also metabolize exogenous drugs and potentially toxic chemicals. The human CYP superfamily consists of 57 genes and more than 59 pseudogenes divided into 18 families and 43 subfamilies. The CYP26 subfamily of enzymes (CYP26A1, CYP26B1/CYP26A2, CYP26C1) are retinoic acid hydroxylases responsible for the inactivation of all-trans-retinoic acid to hydroxylated forms, such as 4-oxo-, 4-OH-, and 18-OH-all-trans-retinoic acid. CYP26B1 is overexpressed in colorectal cancer (CRC) cells, high CYP26B1 expression level in CRC patients is associated with poor prognosis. CYP26B1 mutations are found in radiohumeral fusions with other skeletal and craniofacial anomalies (RHFCA), a disease characterized by craniofacial malformations, occipital encephalocele, radiohumeral fusions, oligodactyly, advanced osseous maturation, and calvarial mineralization defects. |
