Background Information | Cytochrome b-245, beta polypeptide (chronic granulomatous disease), also known as CYBB and P91-PHOX, is a human gene encoding a glycoprotein. Cytochrome b (-245) is composed of cytochrome b alpha (CYBA) and beta (CYBB) chain. It has been proposed as a primary component of the microbicidal oxidase system of phagocytes. CYBB deficiency is one of five described biochemical defects associated with chronic granulomatous disease (CGD). In this disorder, there is decreased activity of phagocyte NADPH oxidase; neutrophils are able to phagocytize bacteria but cannot kill them in the phagocytic vacuoles. |

Immunogen | KLH-conjugated, synthetic peptide corresponding to amino acids 548-560 of human gp91-phox (KQSISNSESGPRG). The immunizing sequence is identical in mouse, and it shares 11/13 sequence homology in bovine and 10/13 in porcine. |
Concentration | Please refer to the Certificate of Analysis for the lot-specific concentration. |
Host | Rabbit |
Specificity | Detects gp91-phox, Mr 75 kDa. Depending on the type of lysate, the amount loaded, the gel and the blotting conditions, gp91-phox may run as a sharp band, Mr 75 kDa, or appear as a smear between 75-91 kDa. |

Entrez Gene Summary | Cytochrome b (-245) is composed of cytochrome b alpha (CYBA) and beta (CYBB) chain. The cause of the killing defect is an inability to increase the cells respiration and consequent failure to deliver activated oxygen into the phagocytic vacuole. |

UniProt Summary | FUNCTION: SwissProt: P04839 # Critical component of the membrane-bound oxidase of phagocytes that generates superoxide. It is the terminal component of a respiratory chain that transfers single electrons from cytoplasmic NADPH across the plasma membrane to molecular oxygen on the exterior. Also functions as a voltage-gated proton channel that mediates the H(+) currents of resting phagocytes. It participates in the regulation of cellular pH and is blocked by zinc. COFACTOR: FAD (Probable). SIZE: 570 amino acids; 65336 Da SUBUNIT: Composed of a heavy chain (beta) and a light chain (alpha). SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein. PTM: Glycosylated. DISEASE: SwissProt: P04839 # Defects in CYBB are a cause of X-linked chronic granulomatous disease (X-CGD) [MIM:306400]. X-CGD is characterized by the failure of activated phagocytes to generate superoxide. Patients suffer from life-threatening bacterial/fungal infections. Patients with CYBB dysfunction are generally classified into three groups, namely X91(0) (no expression of the protein); X91(-) (reduced expression) and X91(+) (normal expression). SIMILARITY: SwissProt: P04839 ## Contains 1 ferric oxidoreductase domain. |
Molecular Weight | Varies. Depending on the type of lysate, the amount loaded, the gel and the blotting conditions, gp91-phox may run as a sharp band, Mr ~75 kDa, or appear as a smear between 75-91 kDa. |

Quality Assurance | Routinely evaluated by Western Blot on human neutrophil membranes. Western Blot Analysis: 1-4 µg/mL of this lot detected gp91-phox in a HL60 membrane and RIPA cell lysate. A previous lot detected gp91-phox in human neutrophil membranes. |

Storage and Shipping Information | |
---|---|
Storage Conditions | Stable for 1 year at -20°C from date of receipt. For maximum recovery of product, centrifuge the vial prior to removing the cap. Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage. |
