Background | Repressor element 1-silencing transcription factor (REST), also known as neural-restrictive silencer factor (NRSF), is a repressive transcription factor that acts by binding to neuron-restrictive silencer elements present in many neuronal genes (1,2). The C-terminal domain of REST is bound by its cofactor, CoREST, which in turn recruits many epigenetic regulators, including HDAC1/2, Sin3A, MeCP2, and LSD1 (3-5). REST is capable of local and long-range chromatin remodeling, including changes to nucleosome phasing and histone modifications (6,7). REST also plays a key role in miRNA regulation along with CREB through the control of genes involved in miRNA biogenesis (8). Many of the miRNAs regulated by REST exist in feedback loops, which control neural stem cell self-renewal and maintenance (9,10). REST also is capable of regulating many long non-coding RNAs (11). REST and its regulatory network of genes and RNAs have been implicated in numerous neurological disorders, including schizophrenia, Huntington’s, and Alzheimer’s disease (12-14). |
