Background Information | Etanercept is a chimeric fusion protein consisting of the extracellular ligand-binding portion of the p75 TNF receptor linked to the Fc portion of human IgG1. This Fc component contains the hinge, CH2 and CH3 regions, but not the CH1 region of IgG1. Etanercept binds to TNF-alpha and blocks its interactions with receptors. Hence, it is considered a TNF inhibitor that functions as a decoy receptor that binds to TNF. Although it mimics the inhibitory effects of naturally occurring soluble TNF receptors, the difference is that Etanercept has a greatly extended half-life in the bloodstream. Its speci fic activity is reported to be about 1.7 x 106 units/mg. Etanercept is shown to modulate biological responses produced by TNF, including expression of adhesion molecules responsible for leukocyte migration, serum levels of cytokines, and serum levels of matrix metalloproteinase-3. It is also shown to affect animal models of inflammation, including murine collagen-induced arthritis. Clone ETA63C8 can bind to Etanercept and block its activity. |
